This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin. Desmoglein 1 is synthesized exclusively in the suprabasal layers. In vertebrates, as in yeast, multiple cyclins have been identified, including a total of eight such regulatory proteins in mammals. Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GIST s), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97.8 percent of scorable GIST s, including all c-kit negative GIST s. Prealbumin, also designated transthyretin, is a major thyroid-hormone binding protein involved in transporting thyroxine from the bloodstream to the brain. Prealbumin is located in the cytoplasm and in the vesicles of developing rat brain cells, and is thought to be transported there from the cerebrospinal fluid via endocytosis. Sequence variants of this protein have been identified in amyloid fibrils from patients with familial amyloidotic polyneuropathy (FAP), the most common form of hereditary systemic amyloidosis. Although the biologically active form of Prealbumin is a tetramer, the amyloidogenic intermediate is thought to be a monomeric species. 8-Oxoguanine DNA Glycosylase (OGG1), otherwise known as N-Glycosylase/DNA lyase, is the enzyme responsible for the excision of 8-oxoguanine (8-oxoG), a mutagenic base by-product which occurs as a result of exposure to reactive oxygen. The action of this enzyme includes lyase activity for chain cleavage. Accumulation of 8-oxoG increases the occurrence of A:T to C:G or G:C to T:A transversion mutations, because 8-oxoG forms a stable basepair with adenine as well as with cytosine. The OGG1 gene encodes eight isoforms (OGG1A-C, OGG2A-E) which result from alternative splicing of a single messenger RNA. Ferritin heavy chains catalyze the first step in iron storage, the oxidationof FeI(II), whereas ferritin light chains promote the nucleation of ferrihydrite, enabling storage of Fe(III). The most prominent role of mammalian ferritins is to provide iron-buffering capacity to cells. Recognizes a protein of 54 kDa, which is identified as cyclin A. Its epitope is located amino acids 144-148 of human Cyclin A2. Cyclins are regulatory subunits of the cyclin-dependent kinases (cdk's) and they control transition at different specific phases of the cell cycle. The temporal expression of cyclins is tightly regulated and subsequently plays a critical role in controlling the enzymatic activity of cdk's. These cyclin/cdk complexes are essential for passage through specific stages in the cell cycle. In mammalian somatic cells, cyclin A is required for S-phase and passage through G2-phase.
